IL-21基因对肝癌细胞抑瘤效应的辐射增敏作用

摘    要:目的:探讨外源性IL-21基因联合放射对肝癌HepG2细胞抑瘤效应的辐射增敏作用。方法:将Ad—IL-21腺病毒颗粒转染HepG2细胞系,进行6Gy”’Cs7射线照射,将细胞分为4组,包括空白对照组、Ad—IL-21组、照射组以及联合组,观察HepG2细胞的生长、细胞周期和细胞凋亡的变化。结果:与Ad—IL-21组和照射组比较,联合组HepG2细胞的抑制效应最显著,转染48h的抑制率明显高于转染24h和72h的抑制率。联合组HepG2细胞阻滞在G2期最多,凋亡细胞所占比例最高,转染24h和48h的凋亡率分别达到29.1%和33.1%。结论:IL-21基因联合放射对肝癌细胞的抑瘤效应具有辐射增敏作用,明显地抑制肝癌细胞的生长。

Abstract:Objective:To study the increased radiosensitivity effect of exogenous interleukin-21 gene on the inhibition growth of hepatic carcinoma cells HepG2. Methods: HepG2 cells line was transfected with Ad- IL-21 and irradiated with 6 Gy 137Cs 3'-ray. The cohorts were divided into blank control, Ad-IL-21, radia- tion, and Ad-IL-21 combined with radiation (combination) groups. The cell growth, cell cycle and cell ap- optosis of HepG2 cells were detected. Results: Compared with Ad-IL-21 and radiation groups, the cell growth in the combination group was inhibited significantly. The inhibition rate was significantly higher af- ter 48 h transfection than after 24 h and 72 h transfection. Cells of G2 phases and apoptosis of HepG2 cells in the combination group were the highest. The cell apoptosis rate in the combination group after 24 h and 48 h transfection reached 29.1% and 33.1% respectively. Conclusion: IL-21 gene combined with ionizing radiation can increase apparently the radiosensitivity effect on the inhibition growth of HepG2 cells.
