| 摘 要： | 目的：以供体造血干细胞（HSC）诱导异基因大鼠肾脏移植免疫耐受,探讨免疫耐受的形成机制。方法：建立大鼠肾脏移植模型,通过免疫磁珠法负筛选系统提取供者骨髓中的造血干细胞。给低剂量放射线全身照射的Wistar大鼠管饲CsA,接着经尾静脉推注SD大鼠的造血干细胞,并当天完成肾移植。观察各组大鼠的存活时间,监测血清肌酐浓度,并进行移植肾彩色多普勒超声。采用体外混合淋巴细胞反应（MLR）法检测长期存活大鼠的免疫耐受状态。结果：实验组大鼠存活时间与对照组比较有显著差异。混合淋巴细胞培养提示,长期存活的受者脾细胞对供者脾细胞的刺激不表现出明显细胞增殖反应,而对无关大鼠脾细胞产生较明显的增殖反应。结论：应用造血干细胞能够成功地诱导出针对供者特异性的免疫耐受。
