摘    要:作者用盐酸胍法从615系小鼠原发性可移植性肝癌(H651、H22)实体瘤组织及正常615系小鼠肝组织(N615)中制备总RNA,经Oligo(dT)纤维素亲和层析法纯化得到 poly(A)~+RNA,再经兔网织红血球溶胞物体系测活及电泳鉴定后,作Northern 印迹和经α-~(32)P-标记的H-ras、Ki-ras及C-myc癌基因分子探针杂交,发现:仅H615中有H-ras基因的明显表达信号,N615中仅有弱表达。

Abstract:Poly (A)~+ RNA from mouse 615 hepatoma(H615, H22) and normal mouse 615 liver(N615) was isolated by guanidinium chlorideand the Oligo (dT) cellulose affinity chro-matography method. RNA was denaturedby the formaldehyde-formamide methodand performed with the agarose electro-phoresis. After the Northern blot transferthe nitrocellulose filters were hybridizedwith ~(32)P-labeled H-ras, N-ras, Ki-ras andC-myc probes. The results showed thatthere was an expression of H-ras in H615,but a very weak expression in N615.
