脂联素基因在3T3-L1脂肪前体细胞诱导分化中表达水平变化的研究

摘    要:
目的:观察脂联素基因mRNA在3T3-L1脂肪前体细胞诱导分化过程中表达水平的变化。方法:体外培养3T3-L1脂肪前体细胞,应用MIX、地塞米松、胰岛素诱导其分化,采用RT-PCR技术检测诱导分化不同时间(0~10天)脂肪细胞中脂联素基因mRNA的表达水平。结果:脂联素基因低表达于3T3-L1脂肪前体细胞中,并随脂肪前体细胞分化成熟,该基因表达水平呈逐渐上调趋势,除在诱导剂作用后第0天与第2天、第1~2天、第3~4天、第6天与第8~10天、第7~10天各时段内差异无显著性(P>0.05)外,其余各时段之间表达水平差异均有显著性(P<0.05)。结论:在3T3-L1脂肪前体细胞分化过程中脂联素基因表达逐渐上调,可能有利于脂肪细胞的分化成熟。

Abstract:
Objective: To observe the change of adiponectin gene mRNA expression during 3T3-L1 preadipocyte differentiation. Methods: 3T3-L1 preadipocytes were cultured in vitro and differentiated into the mature adipocytes induced by 1-methyl-3-isobutylxanthine(MIX), dexamethasone (Dex) and insulin. Adiponectin gene mRNA expression was evaluated by RT-PCR during the process of differentiation at various time points(day 0 to day 10). Results: In preadipocytes,the expression of adiponectin gene mRNA was the lowest. The level of adiponectin gene mRNA was up-regulated during the process of differentiation. There was a significant difference between any two detected time points in the level of the gene mRNA (P < 0.05), except between day 0 to day 2, day 1-2, day 3-4, day 6 and each day of day 8-10, and between any 2 days of day 7-10(P > 0.05). Conclusions: Adiponectin gene mRNA expression is up-regulated during 3T3-L1 preadipocyte differentiation. It may contribute to the adipocyte differentiation and obesity.
