邻苯二甲酸二丁酯对雄性果蝇生殖能力影响及其遗传毒性

摘    要:目的:研究邻苯二甲酸二丁酯(di—n-butyl phthalate,DBP)对雄性果蝇生殖能力的影响及其遗传毒性.方法:①将未交配雄性果蝇随机分为5组,分别给予含DBP为0,1,4,16,64mg/g的培养基喂饲,10d后取存活的实验雄蝇进行交配试验和生育试验.②将雄性果蝇随机分为4组,分别喂饲含DBP为0,50,200,800mg/L的溶液,采用伴性隐性致死(SLRL)试验,观察DBP对雄性果蝇生殖细胞的致突变作用.结果:染毒10d后随着DBP浓度升高各染毒组雄蝇1h交配率和子代发生量逐渐降低,64mg/g组分别为40%和486±265,明显低于对照组的85%和861±312(P〈0.05);生育期1—10d各染毒组的不育率均显著高于对照组(P〈0.01),10d后只有较高浓度组的不育率高于对照组(P〈0.05);50mg/L浓度组精母细胞Ⅲ阶段的SLRL突变率(0.990%)显著高于对照组(0.106%,P〈0.01).结论:DBP对性成熟的雄性果蝇能造成一定的生育障碍和遗传损伤.

Abstract:AIM: To study the effects of dibutyl phthalate (DBP) on the procreant ability and its genotoxity in male Drosophila melanogaster. METHODS:(1)The non-copulated male Drosophila melanogasters were divided randomly into 5 groups and were continuously fed with medium containing 0,1,4,16,64 mg/g DBP for 10 days. Then the treated male flies were brought to the copulation trial and the reproduction trial. (2)The male Drosophila melanogasters were divided randomly into 4 groups and were continuously fed with solution containing 0,50,200,800 mg/L DBP for 3 days. The mutation-inducing effects of DBP was observed using a Sex-Linked Recessive Lethal Test (SLRL test) in Dro- sophila melanogaster. RESULTS:The mating rate in 1 hour and the average reproduction of male flies in DBP groups were gradual- ly decreased with the rising of DBP concentration. The mating rate in 1 hour ( 40% ) and the average reproduction ( 486 ± 265 ) of male flies in 64 mg/g DBP group were lower significantly than those in control group respectively 85% , ( 861 ± 312 ) , P 〈 0.05]. During the first 10 days of the procreation period, the yield rate of male flies in DBP groups were obviously higher as compared with control group ( P 〈 0. 01 ). After continuously procreating for 10 days, the yield rate of male flies merely in higher concentration of DBP groups were significantly higher than that of control group( P 〈 0.05 ). The mutation rates (0. 990% ) of DBP at 50 mg/L concentration in third brood were significantly higher as compared with control group ( 0. 106%, P 〈 0.01 ). CONCLUSION: DBP might reduce the procreant ability and bring out genotoxity effect on male Drosophila melanogaster.
