摘    要:目的探讨实验性肝纤维化基质分解素1基因表达的动态变化。方法建立四氯化碳中毒性及人血白蛋白免疫损伤性两种大鼠肝纤维化模型,采用逆转录定量PCR方法,测定肝纤维化不同阶段基质分解素及基因表达水平。结果正常大鼠肝组织有基质分解素1的基因表达。在肝纤维化的发生、发展过程中,肝组织基质分解素1(MMP_3)基因表达逐渐增强;到肝硬化阶段MMP_3基因表达较肝纤维化阶段下降;在肝硬化的自发逆转过程中MMP_3基因表达保持在明显高于正常对照的水平。结论基质分解素1在肝纤维化的发生、发展和逆转过程中可能起重要作用。

Abstract:Objective To investigate the dynamic alteration of stromelysin-1 (MMP_3) gene expression in the processes of experimental liver fibrosis. Method CCl_4 and albumin induced two animal model ofS liver fibrosis were established and MMP_f gene expression level in the different stages of liver fibrosis was measured by quantitative analysis of reverse transcription polymerase chain reaction (RT-PCR). Result MMP_3 gene expression could be detected in normal rat liver. In the pathogenesis and development of liver fibrosis, MMP_3 gene expression level was increased gradually. At stage of liver cirrhosis, MMP_3 gene expression was lower than that at stage of liver fibrosis. MMP_3 gene expression maintained at relative high level in reversal of liver cirrhosis. Conclusion MMP_3 might play an important role in the processes of pathogenesis, development and reversal of experimental liver fibrosis.
