Abstract:OBJECTIVE To explore the changes of metabolites in sera of patients with end-stage renal disease(ESRD) maintained on hemodialysis using gas chromatography-mass spectrometry(GC/MS) metabolomics to search for potential biomarkers related to ESRD and elucidate the mechanism of ESRD metabolic disorders.METHODS GC/MS was employed for detecting the metabolites in sera of 63 ESRD patients(case group) and 20 matched healthy people(control group).Supervised partial least squares discrimination analysis(PLS-DA) was utilized for analyzing the serum metabolic profiles of samples to search for differential metabolites between two groups.RESULTS The metabolic profiles of control and case group were well-distinguished.And 34 differential metabolites were identified through the databases of NIST,Wiley Registry metabolome and standards.The ROC curve of 34 differential metabolites was plotted and the value under the curve was calculated.Substances with an area greater than 0.5 included 28 metabolites of hydroxylamine,urea and fumaric acid,etc.CONCLUSION ESRD patients have metabolic abnormalities of energy metabolism,amino acid metabolism,lipid metabolism,urea cycle and purine metabolism.Hydroxylamine,urea and fumaric acid may be potential diagnostic markers for ESRD.
