阻塞性黄疸大鼠胃黏膜损害中的细胞信号传导

摘    要:[目的]探讨Toll样受体4(TLR4)介导的信号传导在阻塞性黄疸大鼠模型胃黏膜损害中的作用。[方法]将48只雌性Wistar大鼠随机分为假手术组、对照组、胆总管结扎1d(E1)、3d(E3)、7d(E7)和14d(E14)组,每组8只,胆总管结扎组双重结扎胆总管建立阻塞性黄疸模型(OJ)。用荧光定量逆转录-聚合酶链反应(RT-PCR)对胃黏膜组织细胞中Toll样受体4(TLR4)和肿瘤坏死因子α(TNF-α)mRNA做定量分析。[结果]E1组与E3组间TLR4和TNF-αmRNA的表达差异无统计学意义(P﹥0.05),E7组的表达高于E1、E3、对照组和假手术组(P﹤0.05),E14组的表达高于其余各组(P﹤0.05)。假手术组、对照组、E1、E3组间差异无统计学意义(P﹥0.05)。[结论]阻塞性黄疸时,TLR4介导的信号传导在胃黏膜损害中起了重要作用。

Abstract:[Objective]To study the role of Toll-like receptor4(TLR4)mediated signal transduction in the damage of gastric mucosa with obstructive jaundice(OJ)rat model.[Methods]48 Wistar female rats were randomly assigned to three groups:control group,sham operation group and experiment group.The experiment group was divided into four subgroups according to the days after operation on 1 day,3 day,7 day and 14 day(E1,E3,E7 and E14),there were 8 rats in each group.The experiment groups underwent the common bile duct double bound to form the rat model of obstructive jaundice.Real-time reverse transcription-polymerase chain reaction(RT-PCR)was used to quantitatively determine the expressions of TLR4、TNF-α mRNA in gastric mucosa tissues.[Results]The difference of TLR4 and TNF-α mRNA expression between E1 and E3 was not obvious(P﹥0.05),the expression in E7 was higher than E1,E3,control group and sham group(P﹤0.05),and the expression in E14 was higher than all other groups(P﹤0.05).The expression changes of TLR4 and TNF-α between the control group and sham group was not obvious(P﹥0.05),and the expression changes of these two groups were not significant with E1 and E3 groups(P﹥0.05).[Conclusion]The TLR4 mediated signal transduction plays a role in the damage of gastric mucosa with obstructive jaundice(OJ)rat model.
