摘    要:
目的 观察裉黑素对幼龄、老龄及光制颠倒条件下小鼠交叉上核内c-fos基因蛋白和mRNA水平的影响。方法 采用冰冻切片免疫组织化学染色法,观察裉黑素对小鼠视交叉上核c-fos蛋白水平的影响,采用RT-PCR法,扩增视交叉上核内c-fosmRNA,用琼脂糖凝胶电泳法检测扩增产物。结果 幼禽鼠c-fos蛋白水平及mRNA水平均高于老龄鼠,光制颠倒条件下。c-fos蛋白及c-fosmRNA水平均提高。结论

Abstract:
AIM To observe the c-fosgene expression in the suprachiasmatic nuclear (SCN) of mice after melatonin(MEL)treatment under normal and light-dark shifting circumstances. METHOD c-fos production wasdetermined by the immuno-histochemical staining with the anti-murine c-fos antibody; Aregion selective RNA extraction and rapid RT-PCR method was used to analyse mRNA level inthe SCN. RESULTS The levels of c-fos protein and mRNA in the young micewere higher than those in the old mice. In the light-dark shifting (LD 1410, alternating every 3days), both the levels of c-fos protein and mRNA in each group were decreased. MEL wasfound to enhance their expression. CONCLUSIONS c-fos expression is down regulated along withaging,Light-dark shifting inhibits its expression, MEL(sc) administration can stimulatec-fos expression even under the aging and light-dark shifting conditions, Indicating thatMEL might exert its anti-aging effects through increasing c-fos gene expression.
