丹酚酸B对口腔癌的化学预防作用

摘    要:
目的探讨丹酚酸B对二甲基苯丙蒽(DMBA)诱导的金地鼠颊囊黏膜癌变的化学预防作用及其对血管生成的影响。方法60只雄性金地鼠随机分四组。连续6周(每周3次)在A组(n=20)和B组(n=20)动物右侧颊囊黏膜涂搽DMBA诱发口腔癌,C组(n=10)动物右侧颊囊黏膜涂搽丙酮,D组(n=10)动物不做任何处理,正常饲养。从第7周开始B组和D组动物每天给予丹酚酸B溶液灌胃(10mg/kg),干预癌变过程;A组和C组动物每天给予同等体积的生理盐水灌胃,持续12周。实验结束时处死所有动物,取颊囊黏膜进行组织病理学检查,免疫组化法对血管内皮细胞染色后计数血管密度。结果丹酚酸B能显著降低金地鼠颊囊黏膜癌的发生率(P〈0.01),并能抑制癌变过程中新生血管的形成(P〈0.005)。结论丹酚酸B有望成为一种理想的口腔癌化学预防药物。

Abstract:
Objective To investigate the chemopreventive potential of salvianolic acid B against 7,12-dimethylbenz(a)anthracene(DMBA)-induced hamster buccal pouch carcinogenesis with respect to angiogenesis. Methods(Sixty) male Syrian golden hamsters were randomly divided into 4 groups(group A and B,n=20;group C and D,(n=10).) DMBA solution(0.5% in acetone) was applied topically to the right cheek pouch of male Syrian golden hamsters in group A and B,while animals in group C were painted with acetone,3 times a week for 6 weeks.Animals in group D received neither DMBA nor acetone in the first 6 weeks.Two days after the last treatment of(DMBA,) animals in group B and D received salvianolic acid B daily(10 mg/kg) by gavage,while animals in group A and C received same volume of saline,for the next 12 weeks.At the end of the experiment,animals were sacrificed and tissue samples were collected for histopathological and immunohistochemical examination. Results Salvianolic acid B significantly decreased the incidence of squamous carcinoma(P<0.01).The formation of microvessels was inhibited in papillomas and squamous carcinomas(P<0.005). Conclusion Salvianolic acid B may be a promising chemopreventive agent for oral cancer.
