摘 要: | 目的:通过结扎兔肾动脉分支建立慢性肾衰竭模型,观察主动脉病理变化。方法:新西兰白兔60只,随机分为慢性肾衰竭组(CRF组)和假手术组(Sham组)。CRF组结扎左肾动脉分支同时切除右肾,术后动态观察兔主动脉、残余肾的病理形态学变化和肾功能等指标。结果:CRF组兔术后血清BUN、Scr逐渐升高,肾脏病理逐渐出现明显的肾小球硬化和肾间质纤维化;主动脉逐渐出现内皮细胞脱落、脂纹、粥样斑块等动脉粥样硬化的表现。结论:通过结扎兔左肾外动脉分支和切除右肾一次完成的方法可成功建立CRF加速性动脉粥样硬化模型。

Abstract: | Objective:To establish the rabbit model of chronic renal failure by the branches'deligation of arteria renalis and to study characteristics of pathological changes in the aorta.Methods:60 healthy New Zealand white rabbits were randomly divided into sham-operated group(sham group) and chronic renal failure group(CRF group).The rabbits of CRF group were ligated the branches of left arteria renalis and ectomized the right kidney.Each group was tested at three time points,4 th week,8 th week,12 th week after operation.At each time point 10 rabbits were used.Urine protein,urine occult blood(UOB),haematoglobin(Hb),red blood cell count(RBC),blood urea nitrogen(BUN),serum creatinine(Scr) and systolic blood pressure(SBP) levels were examined.Aortic and renal pathological changes were observed with morphometry.Results:The rabbits of CRF group developed heavy proteinuria,high levels of creatinine,anaemia and heavy hypertension as compared with those in the sham group.The remnant kidneys showed glomerular hypertrophy and extensive glomerular damage consisted of global or segmental sclerosis,tubular dilation,cast formation,tubular atrophy,interstitial fibrosis and lymphocyte infiltration.The typical changes of artherosclerosis were gradually observed in CRF group.Conclusion:By ligating the branches of left kidney artery and ectomizing the right kidney simultaneously the rabbit model of artherosclerosis in chronic renal failure may be established. |
