人卵丘颗粒细胞凋亡与卵子发育潜能的关系

摘    要:目的:探讨人卵子周围卵丘颗粒细胞的凋亡状态与相应卵子发育潜能的关系。方法:收集卵泡浆内单精子注射(ICSI)患者每个卵子周围的卵丘颗粒细胞,流式细胞仪检测卵丘颗粒细胞的凋亡率,研究其凋亡率与卵子的成熟度、受精率及相应胚胎质量的关系。结果:MⅡ期卵子周围卵丘颗粒细胞的凋亡率明显低于GV期、MⅠ期及退变死亡的卵子周围卵丘颗粒细胞的凋亡率(P<0.05);MⅡ期受精卵子的卵丘颗粒细胞凋亡率显著低于未受精的卵子(P<0.01);发育成优质胚胎的MⅡ期卵子周围卵丘颗粒细胞的凋亡率低于发育成评分较低胚胎的卵子的卵丘颗粒细胞(P<0.05)。结论:卵巢刺激周期获取的卵子的成熟状态和发育潜能与相应卵丘颗粒细胞的凋亡程度相关。

Abstract:Objective:To assess the degree of apoptosis in the human cumulus cells and the maturity and fertilization of the corresponding oocyte.Methods:Thirty women undergoing intracytoplasmic sperm injection(ICSI)were analysed,CGCs of every oocyte were collected,the apoptosis of cumulus cells was examined by flow cytometry(FCM).Results:The incidence of apoptosis was significantly higher in cumulus cells from germinal vesicle(GV)stage and metaphaseⅠ(MⅠ) oocytes compared with metaphaseⅡ(MⅡ) oocytes.Nonfertilized MⅡ oocytes showed significantly higher incidence of apoptosis compared with fertilized MⅡ oocytes.There was a correlation between embryo quality and cumulus cells apoptosis variation.Conclusion:A higher degree of apoptosis was seen in cumulus cells from immature oocytes compared with mature oocytes.Apoptosis in cumulus cells impaired the fertilization rate and corresponding embryo quality of MⅡ oocyte after ICSI.
