Abstract:
AIM To explore the expression level of pituitary tumor transforming gene(PTTG) in bladder cancer and its effect on the malignant biological behavior of T24 cells. METHODS The expression of PTTG protein in bladder cancer tissues and adjacent tissues of cancer was detected by immunohistochemistry. The expression level of PTTG protein in bladder cancer tissues of patients with different ages, genders, lesion diameters, lymph node metastasis and clinical stages were compared, and the correlation between PTTG protein level and various indicators was analyzed by Spearman. PTTG siRNA and empty siRNA were respectively transfected into T24 cells.Western blot was used to detect the expression level of PTTG protein in T24 cells in PTTG siRNA group, blank control group and empty siRNA group. Cell proliferation was detected by MTT assay, apoptosis was detected by flow cytometry, and cell invasion was detected by Transwell assay. RESULTS The expression level of PTTG protein in bladder cancer tissue was significantly higher than that in adjacent tissues(P < 0.05). There was no significant difference in the expression level of PTTG protein in bladder cancer tissues of patients with different ages, genders and lesion diameters(P > 0.05). The expression of PTTG protein in patients with lymph node metastasis was higher than that in those without lymph node metastasis, and there were significant differences in PTTG protein expression in patients with different clinical stages and degrees of differentiation(P < 0.05). The expression level of PTTG protein was positively correlated with lymph node metastasis and clinical stage, and negatively correlated with the degree of differentiation(P < 0.05). The expression level of PTTG protein in the PTTG siRNA group was lower than that of the blank control group and the empty siRNA group(P < 0.05). The inhibition rate of cell proliferation in the PTTG siRNA group was higher than that in the empty siRNA group on 2, 3, and 4 days after siRNA transfection(P < 0.05). The apoptosis rate of T24 cells in the PTTG siRNA group was higher than that of the blank control group and the empty siRNA group, and the number of cells penetrating the matrigel was lower than the blank control group and the empty siRNA group(P < 0.05). CONCLUSION PTTG protein is highly expressed in bladder cancer tissues and is positively correlated with lymph node metastasis and clinical staging. Silencing PTTG can inhibit the proliferation and invasion of T24 cells and promote the apoptosis of T24 cells.
