蛋白酶激活受体在心搏骤停后综合征兔肾组织的表达

摘    要:
目的:探讨蛋白酶激活受体(protease-activated receptor,PAR)在心搏骤停后综合征(post-cardiac arrest syndrome,PCAS)兔肾组织的表达及其意义。方法:采用窒息性心搏骤停制备兔心搏骤停后综合征模型(PCAS组),取股静脉血监测器官功能,心肺复苏后48 h,取肾脏组织,采用免疫组织化学技术分别测定兔肾组织PAR-1、PAR-2蛋白表达情况。同时,以假手术组作为对照组。结果:免疫组化结果显示PCAS组、假手术组肾组织PAR-1平均光密度值分别为(1047.67±186.04)、(2278.79±285.77);PCAS组、假手术组肾组织PAR-2平均光密度值分别为(1495.99±133.26)、(2020.85±419.02),PCAS组兔肾脏组织PAR-1、PAR-2蛋白含量均显著减少,两组间比较差异有统计学意义(P<0.05)。结论:PAR-1、PAR-2蛋白表达减少可能与PAR激活有关,并可能与心搏骤停后综合征的多器官功能障碍发生发展相关。

Abstract:
Objective To investigate the expression and significance of protease-activated receptor in nephridial tissue of rab- bits with post-cardiac arrest syndrome(PCAS). Methods The PCAS model was established by using asphyxia-induced car- diac arrest. Femoral vein blood was collected to monitor organ function. After 48 hours, rabbits were sacrificed and nephridial tissues were taken. Then the level of PAR-1 protein was analyzed by immunohistochemical technique and Western blot. Re- suits The mean optical density of PAR-1 was ( 1047.67 ± 186.04) and (2278.79 ±285.77) in PCAS group and sham group, respectively. The mean optical density of PAR-2 were ( 1495.99 ± 133.26) and ( 2020.85 ± 419.02) in PCAS group and sham group, respectively. Compared with sham group, the expressions of PAR-1 and PAR-2 protein were both sig- nificandy down-regulated in PCAS group( P 〈 0.05 ). Conclusion Lower expression of PAR-1 and PAR-2 protein may be associated with the activation of PAR, which may be involved in the development of multiple organ dysfunction in cardiac ar- rest syndrome.
