| 摘 要： | 目的以柯萨奇病毒B3(CVB3)感染体外培养的小鼠心肌细胞,探讨CVB3感染心肌细胞后对心肌细胞周期、心肌细胞凋亡和心肌细胞浆游离钙的影响.方法制备心肌细胞样本,设正常对照组及实验组(CVB3感染心肌细胞),流式细胞仪、钙荧光探针标记分别检测细胞周期、细胞凋亡及心肌细胞浆内游离钙.结果CVB3感染心肌细胞后,可显著抑制心肌细胞的分裂增殖,诱导心肌细胞凋亡;显著增高心肌细胞浆内游离钙浓度和减少细胞外钙含量.结论CVB3感染体外培养的心肌细胞,可能引起心肌细胞凋亡和心肌细胞浆内钙超载,初步实验提示此种变化可能是病毒性心肌炎心脏损伤的重要机制. |

| Abstract: | Objective:The mouse cardiomyoc ytes cultured in vitro were infected wit h Cosackievirus B3(CVB3)for the investi gation of the cell cycle,apoptosis and c ytoplasmic free calcium level in the CVB 3-infected cardiomyocytes. Methods:Cell culture,flow cytometery, calcium fluorescent probing and atom absor ption spectrophotometery Results:The CVB3-infected cardiomyocy tes showed significant inhibition on the division and proliferation,induction of cell apoptosis,apparent increase in the intracellular free calcium and decrease in the extracellular calcium level. Conclusions:Apoptosis and intracellular calci um overload of the CVB3 infected cardio myocytes are the key mechanism underlyin g the cardiac lesion in the case of vira l myocarditis. |
