摘    要:目的采用反相高效液相色谱法建立同时测定马钱子药材及其总碱提取物中马钱子碱和士的宁含量的方法。方法采用Licrospherc18色谱柱(4.6mm×250mm,5μm),流动相为乙腈:0.01mol/mL庚烷磺酸钠与0.02mol/mL磷酸二氢钾等体积混合溶液(用10%磷酸调pH值为2.8)=27:73,检测波长260nm,柱温30℃,流速为1mL/min。结果马钱子碱在0.1—1.0μg、士的宁在0.12~1.2μg范围内线性关系良好(r=1.000),平均回收率分别为99.88%(RSD=1.06%)、100.060(RSD=0.78%)。结论本方法操作可靠、精确,可用于中药马钱子药材及提取物的含量测定。

Abstract:Objective To develop a RP-HPLC method for the determination of Brucine and Strychnine in Semen Strychni and its extractive of total alkaloids. Methods A chromatographic column of Licrospher C18 (4.6mm×250mm, 5 μm) was used with the mobile phase of acetonitrile : 0.01 mol/L sodium heptane sulfonate and 0.02 mol/L potassium dihydrogen phosphate mixed with equal amount {adjusted pH to 2.8 with 10% phosphonic acid)=27 ; 73, detection wavelength at 260 nm, column temperature of 30 ℃ and flow rate of 1 mL/min. Results The calibration curves of Brucine and Strychnine were both in good linearity in the ranges of 0.1-1.0 μg and 0.12-1.2 μg (r=1.000) respectively. The average recovery rates of Brucine and Strychnine were 99.88% (RSD=1.06%) and 100.06% (RSD = 0.78%) respectively. Conclusion The method is realiable and accurate, which can be applied to determination of Brucine and Strychnine in Semen Strychi and its extractive.
