| 摘 要： | 目的:探讨三七皂甙 Rg_1及 Rb_1对脂多糖(lipopolysaccharide,LPS)引起急性肝损害的保护作用。方法:用胶原酶原位灌流法分离肝细胞,用透射电镜观察肝细胞形态学变化,用 Fura-2-AM 测定细胞内游离钙离子浓度。用[~3H]油酸标记的生物膜法测定分泌型磷脂酶 A_2(secretory phospholipase A_2,sPLA_2),用ELISA 法检测前列腺素 E_2(PGE_2)的活性,用 RT-PCR 检测 sPLA_2基因表达。结果:三七皂甙 Rg_1、Rb_1能显著抑制 LPS 所致的肝细胞凋亡与坏死,明显抑制 LPS 引起的胞内 Ca~(2+)超载,抑制 sPLA_2的活性、PGE_2水平和 sPLA_2基因的表达。结论:三七皂甙 Rg_1、Rb_1能显著抑制急性肝损害可能与其对钙的拮抗和对 sPLA_2的抑制作用有关。 |
