长链非编码RNA LOXL1-AS1在胃癌中的表达及其临床意义

目的探讨lncRNA LOXL1-AS1在胃癌中的表达及其临床意义。方法实时荧光PCR(qRT-PCR)检测89对胃癌组织及癌旁组织标本中lncRNA LOXL1-AS1的表达情况,并分析其与各临床病理因素的相关性;利用Kaplan Meier plotter数据库分析不同LOXL1-AS1表达状态下胃癌患者的总生存率差异,探讨LOXL1-AS1表达评估胃癌患者预后的价值。结果胃癌组织LOXL1-AS1表达量显著高于癌旁组织(P<0.05);胃癌组织中LOXL1-AS1表达量与患者肿瘤直径、局部淋巴结转移、远处转移和TNM分期密切相关(P<0.05),但与患者年龄、性别、腹膜转移、肿瘤分化程度等无相关性(P>0.05);Kaplan Meier plotter数据库纳入分析了631例病例样本,其中455例为高表达,176例为低表达。LOXL1-AS1高表达组胃癌患者生存率显著低于LOXL1-AS1低表达组患者,差异有统计学意义(HR=1.52,95%CI:1.17~1.97,P=0.0014)。结论LOXL1-AS1在胃癌中高表达,可能参与了胃癌发生、发展过程;LOXL1-AS1可能作为预测胃癌患者预后评估的指标。

Expression and clinical significance of long non-coding RNA LOXL1-AS1 in gastric cancer

Objective To investigate the expression and clinical significance of lncRNA LOXL1-AS1 in gastric cancer tissues.Methods The expressions of lncRNA LOXL1-AS1 in 89 pairs of gastric cancer tissues and adjacent tissues were detected by quantitive real time PCR(qRT-PCR). The relationship between LOXL1-AS1 and clinical characteristics was analyzed. The overall survival difference of gastric cancer under different LOXL1-AS1 expression status was analyzed by Kaplan Meier plotter. The prognostic value of LOXL1-AS1 expression was explored.Results The expression level of LOXL1-AS1 in gastric cancer tissues was much higher than that in adjacent tissues(P<0.05). The expression level of LOXL1-AS1 was positively associated with tumor diameter, lymph node metastasis, distance metastasis and TNM stage(P<0.05);but it was not correlated with sex, age, peritoneal metastasis and the degree of tissue differentiation(P>0.05). The Kaplan Meier plotter database contained 631 cases samples, including 455 cases with high expression and 176 cases with low expression. Survival analysis revealed that the survival rate of gastric cancer in high expression group was significantly shorter than that in low expression group(HR=1.52, 95% CI: 1.17-1.97, P=0.0014).Conclusion The expression level of LOXL1-AS1 is significantly increased in gastric cancer tissues, suggesting that expression level of LOXL1-AS1 may be involved in the pathogenesis of gastric cancer. LOXL1-AS1 is expected to be an important marker to predict the prognosis of gastric cancer.
