硒化壳聚糖促进体外培养皮肤成纤维细胞增殖

摘    要:目的研究硒化壳聚糖对体外培养的皮肤成纤维细胞增殖的影响。方法用不同剂量(25、50、100、200、400mg/L)硒化壳聚糖作用于成纤维细胞,光镜下观察药物对细胞形态的影响;MTT法、3H-TdR掺入法、细胞生长动力学研究用于检测药物对细胞增殖影响,LDH漏出率检测药物对细胞增殖及损伤的影响,并以等体积细胞培养液处理为对照组。结果各药物浓度组处理细胞后,细胞形态未见明显改变,均可使细胞吸光度值增加,细胞倍增时间缩短,促进表皮细胞对3H-TdR的掺入(P〈0.05),降低LDH漏出率(P〈0.05)。结论硒化壳聚糖对体外培养皮肤成纤维细胞增殖具有促进作用。

The effect of selenium chitosan on the proliferation of cultured skin fibroblasts in vitro

Abstract:Objective To investigate the effects of selenium chitosan on the proliferation of cultured skin fibroblasts in vitro.Methods The cultured skin fibroblasts were treated with selenium chitosan at the concentrations of 25,50,100,200,400mg/L respectively,and the equal volume of media for control group.The changes of skin fibroblasts morphology were observed by light microscope,and the effects on cell proliferation were detected by MTT,3H-TdR infiltration and cell growth curve analysis,and the cell damage was estimated by the leakage rate of LDH.Results As compared with control group,there was no significant difference in the morphology of skin fibroblasts.After treatment with different concentrations of selenium chitosan,the value of absorption degree at 540nm was increased and the double time of cell was shortened.As compared with control group,selenium chitosan could enhance the absorption for 3H-TdR (P0.05),and could decrease the leakage rate of LDH (P0.05).Conclusion Selenium chitosan can promote the proliferation of skin fibroblasts cultured in vitro.
