银杏内酯B对H2O2诱导PC12细胞氧化损伤的影响

摘    要:目的 :探讨银杏内酯B(ginkgolideB)对过氧化氢 (Hydrogenperoxide ,H2 O2 )引起PC12细胞氧化损伤的保护作用。方法 :用噻唑兰 (MTT)检测PC12细胞的存活率 ;乳酸脱氢酶法检测PC12细胞损伤 ;用硫代巴比妥酸法测定细胞脂质过氧化物 ;同时检测细胞内抗氧化酶的活性。结果 :PC12细胞在 5 0~ 30 0 μmol LH2 O2 作用 3h后 ,细胞的死亡率及乳酸脱氢酶 (LDH)释放明显增加 ,抗氧化酶如超氧化物歧化酶 (superoxidedismutase ,SOD)、谷胱甘肽过氧化酶 (glutathioneperoxidase ,GSH Px)活性显著降低 ,而脂质过氧化物生成增多。银杏内酯B 10~ 10 0 μmol L能显著降低细胞死亡率、LDH释放及丙二醛 (MDA)的生成 ,且能明显提高抗氧化酶的活性。结论 :银杏内酯B可拮抗H2 O2 引起的PC12细胞损伤 ,其机理可能与银杏内酯B减少脂质过氧化物的生成 ,提高抗氧化酶的活性有关

Abstract:Objective: To explore the protective effect of Ginkgolide B on hydrogen peroxide induced PC12 cells cytotoxicity. Methods: The viability and insult degree was monitored by MTT and lactate dehydrogenase assays. The content of malondialdehyde (MDA) was measured by a fluorometric assay to indicate the lipid peroxidation. Superoxide dismutase (SOD) and glutathione peroxidase (GSH Px) in PC12 cells were detected. Results: Treatment of PC12 cells with hydrogen peroxide (150 μmol/L) for 3 h caused a great decrease in cell survival (P<0.001 compared with control). Ginkgolide B 10~100 μmol/L dose dependently attenuated the cytotoxic effect of hydrogen peroxide, and also inhibited hydrogen peroxide (150 μmol/L) induced elevation of lipid peroxidation and decline of antioxidant enzyme activities. Conclusion: Ginkgolide B can protect PC12 cells from hydrogen peroxide induced cytotoxicity, which may be pertinent to superoxide scavenging effect and enhancement of antioxidant enzyme activities.
