高脂血症大鼠基底动脉NPY能神经纤维的变化

摘    要:
目的观察高脂血症大鼠基底动脉神经肽Y能神经纤维的变化。方法(1)饲高脂饮食建立高脂血症大鼠模型;(2)卵蛋白-生物素-过氧化物酶(ABC)免疫组化方法观察高脂血症大鼠基底动脉壁神经肽Y能神经纤维的动态变化。结果(1)定性观察:对照组神经肽Y能神经纤维呈环状攀附于血管周围,分布均匀,并见串珠状膨体,其密度无明显变化;实验组饲喂高脂饮食后,纤维即失去原有的环状分布,排列紊乱,出现大量的增生纤维,膨体清晰可见。(2)定量结果:对照组各时期纤维密度无明显改变;高脂组7d即明显增多,纤维密度为70.4±5.5,30d、90d分别为67.6±3.0、65.6±4.8,虽然较7d有所减少,但各时期纤维密度均较对照组增多(P<0.01)。结论高血脂可促使基底动脉神经肽Y能神经纤维增生,这种变化可能与脑动脉粥样硬化的发生、发展有关。

Abstract:
Objective To observe the changes of innervation containing-NPY on the wall of the basilar artery in hyperlipidemic rats. Methods After fed on the hypercholesterol diet, the model of hiperlipidemic rats were built. The innervation of containing-NPY on the basilar arteries was observed by avidin-biotin-peroxidase immunohistochemical technique.Results (1)qualitative observationon: A number of circular innervations with strings of varicosities appeared on the wall of the basialr artery in the control group, the density of them was not different significantly. After the animals had fed on the hypercholesterol diet, the NPY-containing innervations lose their normal construction, the newborn NPY-containing innervations, which also had varicosities, appeared on the basialr artery; (2)quantitative results: In the control group, the density between two of them was not significantly different, however, in the hyperlipidemic group, the density of innervation increased significantly, the density was 70.4±5.5(7d), 67.6±3.0(30d), 65.6±4.8(90d) respectively, compared with the control group( P <0.01). Conclusions Hyperlipidemia should result in the increase of the NPY-containing innervation, which might be associated with the development of atherosclerosis.
