造影多普勒超声显像检测兔肝VX2肿瘤血液供给与肿瘤血管生成相关性的实验研究

摘    要:目的探讨造影多普勒超声检测兔肝VX2肿瘤血流及其与肿瘤血管生成的关系。方法选用肝VX2肿瘤模型兔8只,注射自制声学造影剂后,应用彩色及能量多普勒观察肿瘤病灶周边和内部血流信号的变化、分布和特征;采用免疫组化技术检测肿瘤血管内皮生长因子(VEGF)表达,并进行半定量分级。结果造影后肿瘤内部血流的显示率明显增加,尤以能量多普勒成像(PDI)变化最为明显;免疫组化显示血管内膜生长因子(VEGF)弥漫性表达于癌巢中,表达阳性率52%~85%。统计结果表明,造影能量多普勒显像检测的血管阳性反应面积百分率分级与VEGF表达程度显著相关(r=0.764,P<0.05)。结论应用多普勒超声造影显像技术,有助于评估肿瘤血管生成。

Abstract:Objective To evaluate the correlation between blood perfusion and tumor angiogenesis of hepatic VX2 tumors assessed with contrast Doppler ultrasound imaging.Methods Eight New Zealand rabbits with hepatic VX2 tumors were selected.After administration of self-made echo contrast agent through ear vein,color Doppler flow imaging(CDFI) and power Doppler imaging(PDI) were used to observe the blood perfusion of tumor lesions,while the expression of vascular endothelial growth factor(VEGF) of the tumor lesions were assessed by immunohistochemistry.Results After the injection ofcontrast agent,there were significant improvements in neovascularity of VX2 tumors on CDFI and PDI,especially more pronounced on PDI.The percentage of VEGF expression was 52-85%.There was significant correlation between the graded percentage of positive areas of the blood flow signals assesed by contrasted PDI and the degree of VEGF expressions.Conclusions These results suggested that PDI after the injection of self-made echo contrast may be helpful in evaluating tumor angiogenesis.
