慢性肾衰听功能损害动物模型的制备

摘    要:目的探讨5/6肾切除大鼠听力损害特点及与肾功能损害程度的关系,建立慢性肾衰听功能损害理想的动物模型。方法选用健康白色纯种红目6周龄SD大鼠12只,其中6只行5/6肾切除法制造模型,另设假手术组6只作对照。术后第10周末行听性脑干反应(ABR)及血清肌酐(serumcrealinine,Scr)、尿素氮(blood urea nitro-gen,BUN)测定。结果模型组ABR明显异常,表现为波Ⅰ、Ⅲ、Ⅴ潜伏期延长,Ⅰ-Ⅴ波间期延长(P<0.05或P<0.01),但与肾功能损害程度不具相关性(P>0.05)。结论5/6肾切除大鼠听功能显著受损,是慢性肾衰听功能损害的较理想的动物模型。

Abstract:Objective To observe the character of hearing loss in rats with 5/6 nephrectomy to establish animal model of chronic renal failure(CRF) hearing loss .The correlations were observed between the occurrence rate of inner ear dysfunction,BUN and serum creatinine levels(Scr).Methods Twelve white heathy sprague dawley rats with red eyes were selected,six rats were performed five-sixths nephrectomy to produce CRF model. The others were established a sham group serving as normal control. ABR,BUN and serum creatinine were measured at tenth week after operation.Results The latency of waves I, III, V and I-V interpeak of ABR were significantly longer in the model group comparing with the control group(P<0.05 or P<0.01).No correlations were observed between the occurrence of inner ear dysfunction and BUN and serum creatinine levels(P>0.05).Conclusion The hearing loss is notable in rats with 5/6 nephrectomy,which may be used as an animal model of experimental CRF caasing hearing loss.
