养阴活血方药对糖尿病大鼠肝脏的氧化应激水平及病理改变的影响

摘    要:目的:探讨养阴活血方药对糖尿病大鼠肝脏氧化应激水平及病理改变的影响。方法采用腹腔注射链脲佐菌素(STZ)造糖尿病大鼠模型,挑选造模成功的大鼠,随机分为四组:糖尿病模型组;中药6周组,中药8周组,中药10周组,另外随机选出8只作为正常对照组,造模成功后分别于6、8、10周取材,测定肝组织氧化指标并进行病理观察。结果与正常对照组比较,糖尿病模型组的丙二醛(MDA)的含量明显增加(P〈0.01),超氧化物歧化酶(SOD)活性、谷胱甘肽过氧化物酶(GSH-Px)活性明显降低(P〈0.01)。与糖尿病模型组相比,中药干预组各阶段MDA的含量明显减少(P〈0.05或P〈0.01),中药8周组和中药10周组SOD活性和GSH-Px活性均有显著升高(P〈0.05或P〈0.01),中药6周组虽有升高SOD活性和GSH-Px活性的趋势,但差异无统计学意义(P〉0.05)。病理学观察,模型组肝细胞索及肝小叶结构紊乱,炎症细胞增多,说明肝组织有损伤。与模型组比较,中药干预组各阶段形态学改变均有改善。结论养阴活血方药具有一定的抗氧化能力,抑制氧化应激,对糖尿病大鼠肝脏具有一定的保护作用。

Effects of Nourishing Yin and Promoting Blood Flow Recipe on oxidative stress level and pathologic changes in diabetic rat liver

Abstract:Objective To discuss the effects of Nourishing Yin and Promoting Blood Flow Recipe on oxidative stress level and pathologic changes in diabetic rat liver. Methods Diabetic rats were induced by intraperitoneal streptozotocin (STZ) injection. Successfully-established model rats were selected and randomly divided into four groups: diabetic group, traditional Chinese medicine (TCM) six-week intervention group, TCM eight-week intervention group, TCM ten-week intervention group. In addition, eight normal rats were randomly chosen as normal control group. Rat livers were extracted respectively after six weeks, eight weeks and ten weeks to determine oxidation index and undertake pathologic observation. Results Compared with normal control group, contents of malonaldehyde (MDA) in rat livers of diabetic group rose significantly (P〈0.01), activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) were notably lower than those of normal control group (P〈 0.01); compared with diabetic group, contents of MDA in rat liv-ers of TCM intervention group in each period decreased strikingly (P〈0.05 or P〈0.01), activities of SOD and GSH-Px in TCM eight-week intervention group and TRC ten-week intervention group increased significantly (P〈0.05 or P〈0.01). Rising tendency was shown from activities of SOD and GSH-Px in TCM six-week intervention group, but there was no statistics difference (P〉0.05). By pathology observation, liver cell cords and hepatic lobules structure of diabetic group were chaotic, with inflammatory cells increasing, which indicated that there were injuries in hepatic tissue. Compared with diabetic group, morphology changes were improved in every phase of TCM intervention group. Conclusion Certain anti-oxidative capability was shown to restrain oxidative stress in Nourishing Yi n and Promoting Blood Flow Recipe, which has some protection to diabetic rat liver.
