摘    要:目的:建立一种快速分离黄芩中总黄酮的方法。方法:运用高速逆流色谱(HSCCC)法进行分离,提取溶剂为二氯甲烷-乙醇-水(7:3:1,V/V/V),上相(二氯甲烷)作固定相,下相(乙醇-水)作流动相,流速为10mL·min-1,转速为855r·min-1,进样量为10mL;采用高效液相色谱法测定总黄酮含量。结果:分离得到的总黄酮含量可达到98.8%,1g黄芩能分离得到178mg黄芩苷。结论:该方法简便、易行,可作为黄芩中总黄酮的分离方法。

Abstract:OBJECTIVE:To establish a rapid method for separating total flavones from Scutellaria baicalensis. METHODS:High speed counter-current chromatography(HSCCC) was adopted. The extraction solvent composed of dichloromethane-ethanol-water(7:3:1,V/V/V) ,using upper phase(dichloromethane) as solid phase and lower phase(ethanol-water) as mobile phase at the flow rate of 10 mL·min-1 with revolution speed of 855 r·min-1. The injection volume was 10 mL. The content of total flavones were determined by HPLC. RESULTS:After separation,the content of total flavones reached 98.8%,and 178 mg baicalin was gained from 1 g S. baicalensis. CONCLUSION:The method is simple and easy,and can be used for the separation of the total flavones from S. baicalensis.
