贝那普利联合螺内酯对大鼠心室重构的干预

摘    要:
目的以内皮素(ET)及一氧化氮(NO)为心室重构观察指标研究贝那普利联合螺内酯应用对心室重构大鼠的影响。方法结扎Wistar雄性大鼠腹主动脉建立压力超负荷性心室重构大鼠模型。48只建模后存活大鼠随机分为4组(每组12只):贝那普利联合螺内酯组(贝那普利10mg·kg-1.d-1,螺内酯15mg·kg-1.d-1),贝那普利组(贝那普利20mg·kg-1.d-1),螺内酯组(螺内酯30mg·kg-1.d-1),生理盐水组(生理盐水10ml.kg-1.d-1);不结扎腹主动脉的12只作为假手术组(蒸馏水10ml.kg-1.d-1);各组每天灌胃给药1次,共6周。给药6周后用放免法测定心肌中ET和血浆中NO含量。结果建模组与假手术组相比,心肌中ET含量明显升高(P〈0.05),血浆NO水平明显降低(P〈0.05),表明大鼠心室重构模型建立成功。贝那普利联合螺内酯组及贝那普利、螺内酯单独给药组均能降低心肌ET的含量及升高血浆中NO含量(P〈0.05或P〈0.01),但贝那普利联合螺内酯组比贝那普利组、螺内酯组作用更明显(P〈0.05)。结论联合应用贝那普利与螺内酯可更有效逆转大鼠心室重构。

Abstract:
Objective To investigate the effect of benazepril and spironolactone co-treatment on ventricular remodeling in rats with overloading pressure using endothelium(ET)and nitric oxide(NO)as the observing indexes.Methods The model of pressure loading ventricular remodeling was established through ligating abdominal aorta in Wistar male rats.Forty-eight survival rats after the model set up were randomly divided into 4 groups(n=12 each):benazepril and spironolactone co-treatment group(benazepril 10 mg· kg-1·d-1,spironolactone 15 mg· kg-1·d-1),benazepril group(benazepril 20 mg· kg-1·d-1),spironolactone group(spironolactone 30 mg· kg-1·d-1),normal saline group(normal saline 10 ml· kg-1·d-1) and 12 rats without ligation were as sham group(distilled water 10 ml· kg-1·d-1).Gavage was administered(qd)in every group.After 6 weeks of feeding by gavage,the contents of ET in myocardium and NO in plasma were determined by radioimmunity.Results Comparing with sham group,myocardial ET was markedly elevated,and plasma NO was markedly lowered in model group.These demonstrated that the model of ventricular remodeling was successfully established.Compared with normal salines group,myocardial ET was markedly lowered,and plasma NO was markedly elevated (all P0.05)after treatment in benazepril and spironolactone co-treatment group,benazepril group and spironolactone group,but these beneficial effects were more significant in benazepril and spironolactone co-treatment group compared with other 2 groups(all P0.05). Conclusions The combination of benazepril and spironolactone can effectively inhibit ventricular remodeling than either agent alone in rats.
