FTY720对大鼠心脏移植急性排斥反应的影响

目的观察FTY720对大鼠心脏移植急性排斥反应(AR)的影响并探讨其相关机制。方法利用Cuff技术建立颈部异位心脏移植模型,受体大鼠随机分为对照组和FTY720组,每组12只。FTY720组自术前3 d至术后11 d管饲FTY720(3 mg·kg~(-1)),对照组管饲等量生理盐水。2组均于术后第5天随机选6只动物抽取外周血后留取标本,其余6只观察存活期。苏木素-伊红染色光镜下观察排斥反应级别;流式细胞仪检测外周血CD4~+T淋巴细胞和CD8~+T淋巴细胞。结果FTY720组心脏移植物平均存活时间长于对照组,排斥反应级别低于对照组,CD4~+T淋巴细胞和CD8~+T淋巴细胞阳性率低于对照组,差别均有统计学意义(P<0.05)。结论FTY720能明显减轻大鼠心脏移植AR。

Effect of FTY720 on acute rejection after cardiac transplantation in rats

Objective To study the effect of FTY720 on acute rejection(AR) after cardiac transplantation and investigate the coherent mechanisms.Methods The model of cervical heterotopic cardiac transplantation was established by cuff technique. The recipients were randomly divided into control group and FTY720 group,12 in each group.The FTY720 group were administered with FTY720 orally at a dosage of 3 mg·kg~(-1) for 3 days before grafting to 11 days after grafting;The control group were administered with physiologic saline in the same way.Six recipients in each group were randomly chosen for collecting blood samples on the 5th day after oparation,the other six recipients reserved to observe the survival duration in each group.Cardiac allograft rejection grades were observed under light microscope.CD4~+ T-lymphocytes and CD8~+ T-lymphocyte were detected by flow cytometry.Results The survival durations of the grafts were significantly longer and the rejection degress were significantly lower in FTY720 group than thoes in control group(P<0.05).The positive rates of CD4~+ T-lymphocyte and CD8~+ T-lymphocyte were significantly lower in FTY720 group than those in control group(P<0.05).Conclusion The FTY720 can reduce the acute rejection in rat after heart transplantation.
