摘    要:
目的:建立高效液相色谱法同时测定柴胡中皂苷a和皂苷d含量的方法。方法采用Diamonsil C18(2)柱(250 mm ×4.6 mm,5μm);流动相为水(A)-乙腈(B),梯度洗脱:0~30 min,25%~65%B,平衡时间为3 min,流速为1.0 mL/min,检测波长为210 nm,柱温为35℃,进样量20μL。结果柴胡皂苷a与皂苷d分别在36~192 mg/L(R=0.9993)和36~192 mg/L(R=0.9991)浓度范围内与峰面积呈良好的线性关系。结论该方法简便、快速、具有良好的重现性和稳定性,可用于柴胡药材的质量控制。

Simultaneous determination of saikosaponin a and saikosaponin d in Bupleu-ri Radix by HPLC
Abstract:
Objective A HPLC method was established to determine the content of saikosaponin a and saikosaponin d in Bupleuri Radix.Methods Separation was achieved on Diamonsil C18(2) column (250 mm ×4.6 mm,5 μm). The mobile phase consisted of water(A) and acetonitrile(B) with gradient elution (0~30 min,25%~65%B) at a flow rate of 1.0 mL/min and the equilibration time was 3 min.The detecting wavelength was set at 210 nm and column temperature was maintained at 35 ℃,and injection volume was 20 μL.Results The linear ranges were 36~192μg/mL(R=0.999 3) for saikosaponin a and 36~192 μg/mL(R=0.999 1) for saikosaponin d.Conclusion The method is simple,rapid stable and reproducible ,which can be used for the quality control of Bupleuri radix.
