摘    要:目的:探讨人工关节假体骨溶解模型动物外周血清和关节液中IL-6与骨溶解的相关性。方法:健康成年新西兰兔40只,随机分为实验组三组及对照组,各10只。于动物右股骨髁间窝钻孔,股骨髓腔内置入1枚直径3mm、长15mm的不锈钢圆针。术后6周开始向三个实验组动物右侧膝关节腔内注入聚乙烯悬液0.5mL;对照组注入等量生理盐水作为对照。每2周注射1次,共5次,分别于第16、18、20周处死三组实验动物,第20周处死对照组动物,分别取得标本液。ELISA法检测不同时间段外周血清和关节液中IL-6的含量。结果:在不同时间段,各组外周血清中IL-6含量的差异无显著性,各组关节液中IL-6含量的差异有显著性(P〈0.05),并随着实验时间的延长而增加。结论:在兔骨溶解动物模型中,动物外周血清IL-6水平在骨溶解过程中没有受到影响,而关节液中的IL-6水平随着骨溶解的进展而升高。

Abstract:Objective: To assess the correlation between IL-6 and periprosthetic osteolysis in rabbit experimental study. Methods: Forty rabbits were randomly divided into four groups, 3 experimental groups and 1 control. For each rabbit, a steel nail with 3 mm in diameter and 15 mm in length was put in the medullary cavity of right femur from intercondylar fossa. Six weeks later, the 3 experimental groups were inmected with 0.5 ml fluid containing with polyethylene particles in knee cavity, and the control group was injected with normal saline only, once per two weeks and 5 times totally. The 3 experimental groups were killed after 16, 18, and 20 weeks, resyectively, and the control group was killed after 20 weeks. Specimen fluids were obtained from peripheral circulation and joint cavity. The level of IL-6 in specimen fluids were measured by enzyme-linked immunoabsorbent assay. Results: For different duration, the level of IL-6 in peripheral blood showed no significant difference in every groups (P〉0.05), while the level of IL-6 in joint fluid was different significantly among groups (P〈0.05), and increased significantly with the time of implantation. Conclusion: In rabbit model of periprosthetic osteolysis, serum level of IL-6 is not affected in the process of ostoelysis, on the contrary, the level of IL-6 in joint fluid increases with progress of osteolysis.
