MiR-191在非小细胞肺癌患者血浆中的表达及临床意义

摘    要:
目的 探究非小细胞肺癌(NSCLC)患者血浆中miR-191的表达及其临床意义。方法 选取90例NSCLC患者和60例健康体检者,实时定量PCR(qPCR)法检测手术前NSCLC患者血浆中miR-191的表达,分析miR-191的表达水平与 NSCLC病理指标的相关性。并通过免疫组化检测NSCLC组织中miR-191靶基因凋亡相关蛋白激酶(DAPK1)的表达。结果 NSCLC患者手术前血浆中miR-191表达水平明显高于正常对照组,两组之间的差异具有统计学意义(P<0.01)。miR-191的表达水平与NSCLC患者年龄、性别、病理类型、肿瘤大小及分化程度无关(P>0.05),而与淋巴结转移及TMN分期有关(P<0.01)。miR-191的靶基因DAPK1在NSCLC组织组织表达降低(P<0.01),DAPK1阴性表达患者手术前血浆中miR-191的表达显著高于DAPK1阳性患者(P<0.01)。结论 NSCLC患者手术前血浆中miR-191的表达水平增高,miR-191可能通过负调控DAPK1的表达参与NSCLC的病理过程。

Abstract:
Objective To investigate the expression and clinical significance of miR-191 in plasma of preoperative patients with non-small cell lung cancer(NSCLC).Methods 90 patients with NSCLC and 60 healthy subjects were enrolled.The expression of miR-191 in plasma was detected by real-time quantitative PCR(qPCR).The correlation between miR-191 expression and pathological parameters of preoperative NSCLC patients was analyzed.The expression of DAPK1 as the direct target of miR-191 was detected by immunohistochemistry.Results The expression of miR-191 in plasma of patients with NSCLC was significantly higher than that of the normal control group.The difference between the two groups was statistically significant(P<0.01).The expression level of miR-191 was not correlated with age,gender,pathological type,tumor size and differentiation of NSCLC patients(P>0.05).However,which was associated with lymph node metastasis and TMN stage(P<0.01).The expression of DAPK1 was decreased in cancer tissues(P<0.01),and the expression of miR-191 in DAPK1-negative patients was significantly higher than that in DAPK1-positive patients(P<0.01).Conclusion The expression of miR-191 in plasma of patients with NSCLC increased.MiR-191 may participate in the pathological process of NSCLC by negatively regulating the expression of DAPK1.
