全反式维甲酸对人肝癌SMMC-7721细胞侵袭能力的影响

摘    要: 目的探讨全反式维甲酸(ATRA)对人肝癌SMMC-7721细胞迁移和侵袭的影响及其分子机制。方法划痕愈合和侵袭实验检测ATRA对SMMC-7721细胞迁移与侵袭能力的影响;real-time PCR、Western blotting检测 ATRA作用SMMC-7721细胞后CD147、MMP-2的表达。结果划痕实验结果显示,以10,20,40 μmol·L-1 ATRA处理SMMC-7721细胞24 h后,较未处理组划痕距离明显增加(P <0.05),表明ATRA可抑制SMMC-7721细胞迁移。Transwell实验结果显示,10,20,40 μmol·L-1 ATRA处理SMMC-7721细胞24 h后,穿膜细胞数较未处理组明显减少(P <0.05),表明ATRA可抑制SMMC-7721细胞侵袭能力。real-time PCR结果显示,以10,20,40 μmol·L-1 ATRA处理SMMC-7721细胞24 h后,CD147和 MMP-2 的mRNA表达均明显下调(P <0.05)。Western blotting结果表明,10,20,40μmol·L-1 ATRA处理SMMC-7721细胞24h后,CD147和 MMP-2蛋白表达均明显降低,差异具统计学意义(P <0.05)。结论ATRA可抑制人肝癌SMMC-7721细胞迁移与侵袭能力,其机制可能与下调CD147和MMP-2有关。

Abstract:Objective To investigate the effects of a11-trans retinoic acid (ATRA) on the migration and invasion of human hepatocellular carcinoma SMMC-7721 cells and its molecular mechanism. Methods The effects of ATRA on migration and invasion were examined by scratch healing assay and Transwell assay,and the expressions of mRNA and proteins of CD147 and MMP-2 in treated SMMC-7721 cells were determined by real-time PCR and Western blotting respectively. Results Scratch healing assay revealed that the scuffing distance of SMMC-7721 cells exposed to 10,20,and 40μmol·L-1 ATRA for 24h was significantly increased compared with non-treatment group,indicating that ATRA could inhibit migration of SMMC-7721 cells (P< 0.05). Transwel1 assay showed that the number of SMMC-7721 cells that migrated through the matrigel after being treated by 10,20,and 40 μmol·L-1 ATRA for 24 h was significantly decreased,compared with the non-treatment group,indicating that ATRA could inhibit invasion of SMMC-7721cells (P<0.05).Real-time PCR showed that expression of CD147 and MMP-2 mRNA was down regulated in SMMC-7721 cells treated by 10,20,and 40 μmol·L-1 ATRA for 24 h,respectively.Western Blotting demonstrated that the expression of CD147 and MMP-2 protein was decreased in SMMC-7721 cells exposed to 10,20,and 40 μmol·L-1 ATRA for 24 h. Conclusion ATRA may inhibit the migration and invasion of human hepatocellular carcinoma SMMC-7721 cells,which may be related to the down-regulation of CD147 and MMP-2.
