厚朴提取物长期灌胃对小鼠肾脏的损害作用

摘    要:目的:观察长期服用厚朴甲醇提取物对小鼠肾脏的损害作用。方法:厚朴提取物混悬液0.5ml/鼠(相当于原药材0.25g)给小鼠灌胃,每天1次,于给药后0、1、2、3个月时称体重;用RP—HPLC法测定24h尿、血、肾脏厚朴酚、和厚朴酚量;检查肾功能指标并行肾组织病理学检查。另设生理盐水、空白混悬液对照组。结果:小鼠体重未见明显改变,药物在肾脏排出的半衰期增加1h以上,血清肌酐、尿素氮和补体3及尿蛋白均增加,病理学检查有明显病理改变。结论:长期服用厚朴对小鼠肾脏具有损害作用,肾损害程度与鼠体内厚朴酚、和厚朴酚的浓度呈正相关。

Abstract:Objective: To observe the impairm ent of induced kidney by methanol extract of Magnolia officinalis after single or chronic administra tion in mice. Methods: Methanol extract of Magnolia officinalis 0 5 ml/mouse (equivalent to 0 25 g of Magnolia officinalis) was given intragastrically as a single dose or qd for 1, 2 and 3 month, respectively, with vehicle as the control. Magnolol and h onokiol, the active gradients of Magnolia officinalis, in 24 h urine, serum and kidney were determined by RP-HPLC method. Clinical che mical parameters including serum creatinine(Cr), urea nitrogen(BUN), serum album i n, complement 3(C 3) and 4 (C 4), and urine protein were measured, and patholo gy of kidney was examined after dosing. Results: The excretion h alf-lives of magnolol and honokiol extended by more than 1 h. The contents of C r, BUN, C 3 and urine protein were increased. A significant impairmen t of kidney was shown with pathological changes. Conclusion: Methanol extract of Magnolia officinalis could induce impairment of kidney after chronic administration in mice.
