摘    要:目的研究不同因素对诱导细胞因子诱导的杀伤细胞(cytokine induced killer cells,CIK)体外增殖能力及杀伤活性的影响。为肿瘤CIK细胞治疗提供实验依据。方法细胞计数法及MTT法测定CIK细胞的增殖能力及杀伤活性。结果低年龄组较高年龄组CIK细胞体外增殖能力及杀伤活性强;在无血清培养基条件下培养的CIK细胞的体外增殖能力及杀伤活性较完全培养基的强;在一定的培养时间内CIK细胞的体外增殖能力与培养的时间成正比;CIK细胞对各种肿瘤细胞均有杀伤作用。结论不同因素对肿瘤患者CIK细胞体外增殖能力及杀伤活性有影响。

Abstract:Objective] To approach and evaluate various effects of different factors on proliferation capacity and cytotoxocity of induced CIK (cytokine induced killer) cells in vitro, and to provide experimental evidence for cell therapy of CIK in tumour. Method] Cytometry and MTT assay were applied in detecting proliferation capacity and cytotoxicity of CIK. Result] The proliferation capacity and cytotoxicity of CIK in vitro appeared stronger in the group of low age or serum free medium than in the group of advanved age or serum medium. And the proliferation of CIK in vitro increased positively with time during a certain period of incubation. Furthermore, CIK had equal cytotoxicity to various tumour cells. Conclusion] Various factors might influence the proliferation capacity and cytotoxicity of CIK of patients with tumour in vitro.
