摘 要: | 目的研究瓜蒌皮提取物(EPT)对氧化低密度脂蛋白(OX-LDL)诱导的人脐带静脉内皮细胞(HUVECs)损伤的保护作用。方法在培养的人脐带静脉内皮细胞中加入OX-LDL(100mg/L)诱导内皮细胞损伤。检测细胞培养液中非对称性二甲基精氨酸(ADMA)、丙二醛(MDA)、乳酸脱氢酶(LDH)、一氧化氮(NO)、二甲基精氨酸二甲胺水解酶(DDAH)和肿瘤坏死因子-α(TNF-α)的含量。结果体外给予OX-LDL(100mg/L)显著诱导内皮细胞损伤,提高培养液中ADMA、MDA、LDH、和TNF-α水平,降低NO和DDAH水平。两种洗脱(水和70%乙醇洗脱)瓜蒌皮提取物均能显著抑制OX—LDL所致的ADMA、MDA、LDH和TNF-α浓度升高和NO、DDAH降低。结论瓜蒌皮提取物对OX—LDL诱导的人脐带静脉血管内皮细胞损伤有保护作用,其保护作用与降低ADMA和TNF-α浓度有关。

Abstract: | Objective To study the protective effect of EPT on damages of the human umbilicus vein endothelial cells (HUVECs) induced by oxidized low-density lipoprotein (ox-LI)L). Methods The injury of endothelial cells was induced by ox-LDL (100mg/L) in cultured endothelial cells. The medium samples were collected from cultured endothelial cells. And the concentrations of asymmetric dimethylarginine (ADMA), malondi- aldehyde (MDA), lactate dehydrogenase (LDH), nitric oxide (NO), dimethylarginine dimethylaminohydrolase (DDAH) and turnout necrosis factor-α (TNF-α) were measured. Results Ox-LDL (100 mg/L) significantly increased the concentrations of ADMA, MDA, LDH and TNF-α, and decreased the concentrations of NO and DDAH. EPT with the different elution (water and 70% alcohol) markedly reduced the injury of endothelial cells by ox-LDL, and inhibited the increase of ADMA, MDA, LDH and TNF-α, and inhibited the decrease of NO and DDAH by ox-LDL. Conclusion EPT protects the endothelial cells against damages induced by ox-LDL, and the effects is related to the reduction of ADMA and TNF-α concentration in cultured endothelial cells. |
