摘    要:
目的:探讨8-溴-7-甲氧基白杨素(BrMC)诱导人非小细胞肺癌A549细胞凋亡作用及分子机制。方法:流式细胞术(PI染色)分别检测白杨素及BrMC处理后的细胞凋亡。蛋白印迹法检测BrMC对Bim蛋白表达以及N-乙酰半胱氨酸对BrMC介导Bim表达的影响。siRNA沉默Bim基因后流式细胞仪检测BrMC对细胞凋亡影响。结果:BrMC对A549细胞的凋亡作用较白杨素更强。经BrMC(10μmol/L)处理后,Bim蛋白水平呈时间依赖性增加。siRNA干扰后,BrMC诱导的A549细胞凋亡率明显降低。结论:BrMC通过上调Bim诱导肺癌A549细胞的凋亡。

Abstract:
Objective To explore the molecular mechanism of 8-bromo-7-methoxychrysin induced apoptosis in human non-small-cell A549 cells.Methods Apoptosis of A549 cells treated with Chrysin and BrMC was measured by Flow cytometry using PI staining.The effect of BrMC on Bim expression in A549 cells or cells transfected with Bim siRNA was analyzed by Western blot.The role of NAC in BrMC-mediated the expression of Bim was determined by Western blot.Results BrMC had more potent effect on apoptosis in A549 cells compared with Chrysin.Treatment of A549 cells with BrMC(10 μmol/L) caused a time-dependent increase in Bim protein expression.Cells transfected with siRNA specific to Bim displayed a reduced proportion of apoptotic death cells induced by BrMC.In addition,pretreatment with NAC(10 mmol/L) inhibited BrMC-mediated Bim expression.Conclusion Our results suggest that BrMC-induced apoptosis of A549 cells was associated with upregulation of Bim protein expression.
