Abstract:Objective To study the effect of interleukin 12 (IL 12) on the therapeutic value of DNA vaccine as well as its tumorigenicity of SP2/0 C transfected with core (HCV C) gene. Methods The HCV core (HCV C) gene cut from pBRTM HCV1 3011 was inserted into the eukaryotic expressive plasmid pcDNA3. Then the recombinant plasmid pcDNA HCV C was constructed and expressed using Lipofectamine in SP2/0 cells. Detection of C protein was proved by immunofluorescence (IF) test. Lastly, mice were immunized by intradermal injections of SP2/0 cells transfected with HCV C gene. Results The tumorigenicity of SP2/0 HCV C cells was reduced significantly by pcDNA HCV C inoculation as compared with mock vector transfected with SP2/0 cells. Moreover, the survival time of mice inoculated with pcDNA HCV C plasmid combined with IL 12 expressive plasmid was prolonged. Conclusion pcDNA HCV C DNA vaccine combined with IL 12 expressive plasmid might get better therapeutic value in the gene therapy against HCV infection.
