摘    要:目的:探讨去甲二氢愈创木酸(NDGA)诱导恶性胶质瘤细胞分化过程中中对GFAP基因表达的影响及其意义。方法:用免疫组织化学和原位杂交方法定性、定量检测人恶性胶质瘤细胞系SHG-44细胞GFAP基因的表达水平。结果:NDGA处理后,瘤细胞GFAP蛋白及mRNA表达水平明显增高(P<0.01)。结论:NDGA具有诱导恶性胶质瘤细胞GFAP基因表达的作用,推测GFAP基因表达上调可能是NDGA诱导胶质瘤细胞分化的作用机制之一。

Abstract:Objective To investigate the expression of glial fibrillary acidic protein (GFAP) gene and its significance in the process of glioma cell differentiation induced by nordihydroguaiaretic acid (NDGA). Methods Immunohistochemistry (IHC) and in situ hybridization were used to detect the expression changes of GFAP protein and GFAP mRNA qualitatively and quantitatively. Results The expression levels of GFAP protein and GFAP mRNA in NDGA treatment group were significantly increased compared with the control group ( P <0.01). Conclusion NDGA could induce GFAP gene in malignant glioma cells and the up regulation of this gene expression might be one of the mechanisms by which NDGA induces glioma differentiation.
