CT-1mRNA和gp130在高血压大鼠心肌中的表达变化及意义

目的观察CT-1mRNA和gp130在高血压大鼠心肌中的表达变化、意义,以及替米沙坦对心肌营养素-1(CT-1)和糖蛋白130(gp130)表达的影响。方法 20只雄性SD大鼠行腹主动脉缩窄术后2周造成压力负荷性心肌肥厚模型,随机分为心肌肥厚组和替米沙坦组,另设10只作为假手术组。替米沙坦组灌胃给药3 mg/(kg.d),连续4周。原位杂交法检测心肌的CT-1mRNA表达水平;免疫组化法检测心肌中gp130表达水平。结果与假手术组比较,心肌肥厚组心肌组织CT-1mRNA和gp130表达水平明显增加。与心肌肥厚组比较,替米沙坦组CT-1mRNA和gp130含量显著降低(P<0.01)。结论压力超负荷性大鼠心肌中CT-1mRNA及其受体gp130的过度表达与心室重构的发生密切相关;替米沙坦逆转心室重构的机制可能与抑制CT-1及受体gp130的过度表达相关。

Changes of expression of CT-1mRNA and gp130 in myocardium and Significance in hypertensive rats

Objective To determine the changes of expression of CT-lmRNA and GP130 in myocardium and significance in hypertensive rats. Methods 20 male SD rats were randomly divided into 2 groups at two weeks after partial ligating aorta： hypertrophied group（LVH, n=10）and telmisartan group（Tel, n=10）.Sham-operated group（Sham, n=10）was selected to serve non-partial ligating aorta. Telmisartan group were delivered by direct gastric gavage [（3 mg/ （kg·d））for 4 weeks. Ang Ⅱ level in myocarcl,ium was measured by radioimmunoassay. The expression of CT-lmRNA of cardiomyocytes was determined with in site hybridization histochemistry. The protein level of gpl30 of cardiomyocytes was determined by immunohistoehemistry analysis..Results Compared with sham-operated group, Cardiotrophin-lmRNA gene expression and the protein expressing of gpl30 in cardiac tissue were significant enhanced （P〈0.01）in the LVH. Conclusion Telmisartan down-regulated the cardiotrophin-1 mRNA and the protein expression of gp 130 in the elmisartan roup.
