中国东北地区HTLV-1的前病毒DNA测定及其意义

目的 了解我国东北的HTLV-1的流行情况及病毒特征。方法 从2002年起在东北地区共采取了941份标本。用免疫荧光法进行了血清抗体的检测，经PCR扩增及Southern杂交鉴定。结果 发现3例HTLV-1阳性感染病人，阳性率0．3％，没有发现HTLV-2阳性感染病人；在37例淋巴细胞白血病患者中检出2例，检出率5．4％。并对其中1例阳性感染病人的外周血单个核细胞（PBMCs）中的前病毒DNA的env基因的gp21、gp46两个碱基对核甘酸序列进行了PCR扩增，并将其克隆到T—Vector上进行DNA的核甘酸序列测定，与日本原型（ATK）病毒DNA链的相应区域的线性比较显示了很高的同源性（98．9％-100．％），没有发现碱基的插入、删除和其他显著性的变化。结论东北地区为HTLV-1的非流行区，于淋巴细胞白血病病人检出率（5．4％）较高，说明东北地区的淋巴细胞白血病与HTLV-1感染有一定的关系。

Determination and significance of retrovirus DNA of HTLV - 1 in northeast China region

High prevalences of HTLV - 1 infection has been found in Japan and other areas To gain the epidemiological presence and viral characterza-tion. Methods Antibodies to HTLV - 1/2 were investigated 941 individuals living in Eeast- North of China. The blood from patients with vinous neurological diseases, leukemia and health person. Antibodies were determined by an inmunoflurescence method, and positive reactions were confirmed by PCR and southern hybridization for the pol gene of the proviral DNA. Results The presence of HTLV - 2 in 941 samples (0 % ), THE PRESCENE Of HTLV -1(0.3%). Conclusion The HTLV - 1 strauns for Esast - Nonth of China were 98.9% idental to Japaues prototype ALK.
