| 摘 要： | 目的利用RNA干扰技术抑制VEGF基因表达，探讨RNA干扰对胃癌细胞的增殖的影响。方法设计靶向VEGF基因shRNA．构建质粒pcDNA3．1-shRNA—VEGF并利用lipofectamine^TM2000转染胃癌细胞株BGC-823．采用RT—PCR和Western blot技术观察胃癌细胞VEGF mRNA及蛋白表达的变化，同时观察胃癌细胞增殖的变化。结果和对照组相比，VEGF—shRNAB、C组VEGF表达和细胞增殖均明显下调。结论RNAi明显抑制了VEGF的表达和胃癌细胞增殖。

| Abstract: | Objective To investigate the effect of small interfering RNA (siRNA) on the inbibiting gene expression of vascular endothelial growth factor (VEGF) as well as the proliferation of gastric cancer cell line BGC-823.
