补肾中药有效成分对SAMP8小鼠海马APP及PS1基因表达的影响

摘    要:目的]观察补肾中药有效成分淫羊藿苷、补骨脂素、齐墩果酸对6月龄SAMP8小鼠淀粉样前体蛋白(APP)及早老素-1(PS1)基因表达的作用,为补肾中药防治老年性痴呆提供实验依据。方法]选择6月龄健康雄性SMAP8小鼠40只,随机分为模型组、淫羊藿苷组、补骨脂素组、齐墩果酸组,每组10只。另取10只SAMR1小鼠作为对照组。治疗组分别给予淫羊藿苷、补骨脂素、齐墩果酸灌胃治疗(浓度为淫羊藿苷20 mg/kg,齐墩果酸、补骨脂素各50 mg/kg),每日1次,每次0.3 mL,连续灌胃4周。对照组与模型组均给予等量生理盐水灌胃。4周后应用逆转录-聚合酶链反应(RT-PCR)方法检测海马组织PS1基因表达。结果]1)与对照组相比,淫羊藿苷组、齐墩果酸组及补骨脂素组对APP基因下调均具有统计学意义(P<0.05)。2)与对照组相比,补骨脂素组有下调小鼠海马组织PS1基因的趋势,淫羊藿苷组及齐墩果酸组对PS1基因下调具有统计学意义(P<0.05)。结论]补肾中药可能通过下调APP及PS1基因的表达,进而达到治疗老年性痴呆作用。

Abstract:Objective] To observe the effects of icariin, psoralen and oleanolic acid on the APP gene and PS1 gene expression in SAMP8 mice of 6 months. To provide experimental basis for prevention and treatment by TCM of reinforcing kidney. Methods] Forty SAMP8 male mice were randomly divided into model group, icariin group, psoralen group and oleanolic acid group taking ten SAMR1 male mice as control. The treatment groups were intragastrically administered with icariin, psoralen and oleanolic acid(icarrin at dose of 50 mg/(kg·d), psoralen at dose of 20 mg/(kg·d) and oleanolic acid at dose of 20 mg/(kg·d) ), 0.3 mL, once a day for 4 weeks. Model group and control group were all intragastrically administered with equivalent saline. After 4 weeks, the hippocampus APP gene and PS1 gene expression were detected by RT-PCR. Results] 1)Compared with control group, the APP gene expression were reduced with statistical significance (P<0.05) in icariin group, oleanolic acid group and psoralen group. 2)PS1 gene expression had reduced trend in psoralen group. Especially, icariin and oleanolic acid could reduce the PS1 gene expression with statistical significance (P<0.05). Conclusion] Active components of TCM with action of reinforcing kidney can be used for treating Alzheimer's disease by reducing the expression of APP gene and PS1 gene.
