LETMD1单克隆抗体的鉴定及其在肝细胞癌临床诊断研究中的初步应用

目的:对制备的LETMD1-C末端单克隆抗体进行免疫特性鉴定,并将其应用于肝细胞癌的临床诊断研究。方法:使用ELISA,细胞免疫荧光及Western blot等免疫学技术对所制备的单克隆抗体进行免疫特性鉴定;以鉴定的抗体为工具,检测肝癌组织中LETMD1编码产物表达情况。结果:免疫学鉴定结果验证了制备的单克隆抗体具有较高的使用效价和良好的特异性。检测肝组织标本中LETMD1编码产物表达情况,发现其在正常组织中不表达,在肝癌组织表达明显高于癌旁组织。结论:制备了抗-LETMD1-C末端的单克隆抗体,经鉴定具有较高的效价和良好的特异性,可以应用于LETMD1癌基因的功能研究和肝细胞癌的临床诊断研究,为深入研究LETMD1蛋白的生物学功能和建立新的肝细胞癌早期诊断方法奠定了可靠基础。

Identification of anti-LETMD1 McAbs and its application in the clinical diagnosis of hepatocellular carcinoma

Objective:To identify immune features of the anti-LETMD1-C McAbs and apply it to the clinical diagnosis in hepatocellular carcinoma.Methods: ELISA,Western blot and immunofluorescence were used to identify McAbs;The identified McAbs were used to detect the LETMD1 protein in Liver tissue specimens.Results: Anti-LETMD1 McAbs were identified by ELISA,Immunofluorescence and Western blot,which revealed that the McAbs can recognize LETMD1-protein specifically with high valence.The identified McAbs were used to detect the LETMD1 protein in Liver tissue specimens,it was found that the expression of LETMD1 in Hepatocellular carcinoma significantly higher than that in para-carcinoma tissue.Conclusion: Anti-LETMD1-C McAbs were successfully identified,they had both high sensitivity and high specificity,the results may provide an experimental foundation for in-depth study on LETMD1 biological functions and establish the sensitive and specific method for early diagnosis of hepatocellular carcinoma.
