中医药治疗老年肿瘤的机理研究

摘    要:目的:探讨中药抗肿瘤疗效及Fas/FasL基因在医疗中的作用。方法:将0、0.01、0.1、1μg/mL表阿霉素分别处理的肝癌细胞与有、无参芪处理的淋巴细胞一起培养,观察中药或FasL单克隆抗体介入之后抗肿瘤的实际效果;用PCR进行测定化疗药物处理的肝癌细胞Fas/FasL基因的具体表达。结果 1μg/mL表阿霉素所处理的肝癌细胞OD值要高于0、0.01、0.1μg/mL表阿霉素所处理的肝癌细胞,没有处理肝癌细胞的OD值则最高;7402肝癌细胞表达Fas和FasL基因,与0μg/mL表阿霉素干预过的7402肝癌细胞FasL表达比较,0.01、0.1μg/mL的降低,1μg/mL的增高;FasL单克隆抗体介入之后,0、0.01、0.1μg/mL表阿霉素处理的肝癌细胞OD值也高于空白对照。结论:中药抗肿瘤可减毒增效;Fas/FasL系统在抗肿瘤免疫机制中起重要作用。

Abstract:Objective:To study the functions of traditional Chinese medicine in the treatment of tumours and the effetiveness of Fas/FasL genes.Methods:Culture the lung cancer cells disposed both by 0、0.01、0.1、1μg/mL pharmoribicin and lymphocyte with and without astragalus and then observe the actual effectiveness of tumours treatment with traditional medicine or FasL monoclonal antibody;to get the actual explanation of lung cancer cell Fas/FasL genes by means of PCR testing.Result:The OD of lung cancer cell with 1μg/mL pharmoribicin is higher than that with 0、0.01、0.1μg/mL pharmoribicin and the highest lies in that without pharmoribicin.Compared Fas amd FasL genes in 7402 lung cancer cells and those disposed in 0μg/mL pharmoribicin,0.01、0.1μg/mL decreased while 1μg/mL increased;after the introduction of FasL monoclonal antibody,the OD in lung cancer cells disposed by 0、0.01、0.1μg/mL pharmoribicin is also higher than the original.Conclusion:Traditional Chinese medicine can reduce the toxicity in tumour treatment and Fas/FasL plays an important role in anti-tumour immune mechanism.
