重楼皂苷I对高转移人卵巢癌细胞体外生长抑制功能的研究

摘    要:目的:研究重楼提取的活性成分重楼皂苷I对高转移人卵巢癌细胞的体外生长抑制及促凋亡功能,并初步探讨其可能的作用机制。方法:用CCK8法检测不同浓度重楼皂苷I在不同时间点对高转移人卵巢癌细胞(HO-8910PM)体外增殖的影响;光镜下观察细胞形态的变化;用流式细胞术(FCM)检测经不同浓度重楼皂苷I处理后高转移人卵巢癌细胞的细胞周期变化;用AnnexinV/PI双染法、Hoechst33258检测不同浓度重楼皂苷I诱导细胞凋亡的发生。结果:重楼皂苷I在0.5~5μg/mL浓度范围对HO-8910PM细胞的增殖均有明显抑制作用,并呈现明显的剂量-时间依赖性。其半数抑制浓度在24h到120h范围内基本集中在1~3.5μg/mL之间;重楼皂苷I主要影响HO-8910PM细胞的DNA合成前期(G0/G1);重楼皂苷I浓度控制在2μg/mL以内,早期即3h开始就能诱导HO-8910PM细胞的凋亡,作用时间越长,凋亡发生率越高。结论:重楼皂苷I能抑制卵巢癌细胞的增殖并诱导其凋亡,从而发挥抗肿瘤作用。

Abstract:Objective:To investigate the proliferation inhibitory and apoptosis effect of Paris Saponin I(PSI),an active component isolated from Rhizoma Paridis,on high metastatic ovarian cancer cell line HO-8910PM in vitro,and explore the possible mechanisms.Methods:We dealed with the cell line HO-8910PM at different times with different concentrations of PSI.CCK8 assay was used to examine the status of cell growth.Changes in cell morphology were observed under optical microscope.Flow cytometry,AnnexinV/PI double-staining and Hoechst33258 methods were adopted to determine cell cycle and apoptosis changes.Results:Different concentrations of PSI(0.5~5μg/mL)all showed markedly inhibitory effect in a dose-dependent manner on HO-8910PM,especially in G0/G1 phase.The concentrations to inhibit cell growth by 50%(IC50)were 1~3.5μg/mL from 24 to 120h.Early use of PSI(at the beginning of 3h,<2μg/mL)could induce apoptosis.Conclusion:PSI can inhibit the metastatic ovarian cancer cell line growth,induce apoptosis,and thereby resist carcinoma.
