碘与海藻玉壶汤对碘缺乏致甲状腺肿干预机制的比较研究

摘    要:目的:从抗氧化应激角度比较碘与海藻玉壶汤对碘缺乏甲状腺肿的干预机制。方法:选4周龄Wist-ar大鼠,制成碘缺乏动物甲状腺肿模型。随机分成4组:除正常对照组(NC)饲正常饲料外,其余组饲以低碘饲料。NC组和模型对照组(MC)每日灌服等体积双蒸水。单纯高碘组(PHI)灌服含碘1900μg/L的高碘水,海藻玉壶汤(HZ)每日灌服按与人相同体表面积折算的海藻玉壶汤。在给予处理因素3周和11周后处死大鼠。采用生化方法测定大鼠血清内谷胱甘肽过氧化酶(GSH-Px)、黄嘌呤氧化酶(XOD)和超氧化物歧化酶(SOD)的活力,以及丙二醛(MDA)及过氧化氢(H2O2)的含量。采用免疫组化方法检测甲状腺中4-羟基壬烯醛(4-HNE)的表达。采用Western印迹及免疫组化技术对过氧化物氧化还原酶5(PRDX5)在大鼠甲状腺的表达及分布进行了分析。结果:用药11周后,HZ组SOD活力呈现上升趋势,明显高于NC组和PHI组(P0.05或P0.001)。HZ组11周较3周时GSH-PX活性有所增加。11周时PHI组H2O2含量明显高于NC组,并且MDA含量明显高于其他组(均P0.05)。11周时XOD活性PHI组和HZ组均明显高于NC组和MC组(P0.001或P0.01)。甲状腺4-HEN表达PHI组明显高于MC组(P0.05),HZ组明显低于NC组和PHI组(P0.05)。甲状腺PRDX5表达免疫组化和Western blot结果均显示HZ组较NC和PHI组显著增高(P0.05)。结论:海藻玉壶汤通过抗氧化应激,可使甲状腺肿恢复更完全,并未造成甲状腺细胞的损伤。

Abstract:Objective: To Compare the intervention mechanisms of iodine and Haizao Yuhu Decoction for goite due to iodine deficiency from the perspective of anti-oxidative stress.Method:4-week-old Wistar rats were made animal model of iodine deficiency goiter and randomly divided into four groups.In addition to the NC group was fed with normal diet,the other groups were fed with a low iodine diet.NC group and MC group were daily fed with an equal volume of double-distilled water.PHI group was fed with potassium iodate solution with the iodine concentration of 1900μg/L.HZ group was daily fed by the same body surface area with human conversion of Haizao Yuhu Decoction.3 weeks and 11 weeks after treatment,rats were killed.Rat serum GSH-Px、XOD and SOD vitality,MDA and H2O2 content were determined by biochemical methods.Immunohistochemical method was used to detect thyroid 4-HNE expression.PRDX5 expression in rat thyroid and distribution were analyzed by Western blot and immunohistochemical.Result:11 weeks after treatment,SOD activity in HZ group was significantly higher than that in the NC group and the PHI group(P<0.05 or P<0.001).Compared with that after 3 weeks of treatment,GSH-PX activity in HZ group of 11 weeks after treatment was increased.H2O2 content in PHI group was significantly higher than that in NC group,and MDA content was significantly higher than that in the other groups(all P<0.05).XOD activity in PHI group and HZ group of 11 weeks were significantly higher than that in NC group and MC group(P<0.001 or P<0.01).Thyroid 4-HEN in PHI group was significantly higher than the expression of MC group(P<0.05).Thyroid 4-HEN in HZ group was significantly lower than that in NC group and PHI group(P<0.05).Expression of thyroid PRDX5 by Immunohistochemistry and Western blot results was significantly higher compared with NC,PHI,HZ groups(P<0.05).Conclusion:Haizao Yuhu Decoction can completely recover Goiter by oxidative stress and dose not cause damage to thyroid cells.
