电针对神经干细胞移植痴呆大鼠学习能力和组织内乙酰胆碱酯酶的影响

摘    要:目的:观察电针对神经干细胞(NSCs)海马内移植老年性痴呆(AD)大鼠学习和组织内乙酰胆碱酯酶含量的影响,探讨电针治疗老年性痴呆的机制。方法:采用β-淀粉样蛋白(β-AP)向海马CA1区定向注射制做AD模型,将大鼠分为5组:正常组、模型组、移植组、电针组和移植电针组,每组各12只。针刺选取百会、大椎、人中3个穴位。大鼠跳台检测学习能力;酶免法检测血清、海马和大脑皮层组织内乙酰胆碱酯酶(AChE)含量。结果:移植电针组大鼠跳台潜伏期比模型组增加(P0.05),错误次数比模型组减少(P0.05);血清、海马和大脑皮层组织内AchE含量比模型组降低(P0.05),血清和海马内AchE含量与移植组比较有显著性差异(P0.05)。结论:电针对大鼠组织内乙酰胆碱酯酶含量的改善是治疗AD的作用机制之一,电针结合神经干细胞移植治疗效果更佳。

Abstract:Objective:To observe the effect of electric needle on the study ability and the quantity of AchE system in brain of AD rats of neural stem cell transplantation of hippocampus and discuss the mechanism.Methods:The model was made by injecting β-AP to CA1 of hippocampus.The rats were randomized into normal,model,transplantal,electric needle and transplantal needle groups,each group had 12 rats.Baihui,Dazhui and Renzhong were selected and connected the electronic needle device.The memorizing ability was measured by the diving tower of rats and enzyme-immunossay methods in cerebral cortex,the content of hippocamous AchE were examined.Result:The incubation of step-jumping of the transplantal needle increased more(P<0.05)than the model group and the correct times decreased more(P<0.05)than the modal group;the quantity of AchE in cerebral cortex and hippocampus decreased more(P<0.05)than the model group;there were obvious different between the transplantal needle and the transplantal groups(P<0.05).Conclusion:The quantity of AchE system in brain of AD rats can be improved by acupuncturing and electric needling,which will be better for transplant of neural stem cell.
