血塞通对痛风炎症因子ICAM-1表达的影响

摘    要:目的:血塞通的主要成分为三七总皂苷(PNS),本实验将PNS对以尿酸钠(MSU)100μg/mL诱导体外急性炎症模型,采用细胞活力以及炎症因子的表达高低来观察血塞通对体外急性炎症模型的影响。方法:细胞分为空白组、刺激祖、给药组和阳性药对照组,采用MTT法测量细胞活性、实时定量PCR检测ICAM-1的基因表达。结果:血塞通2.5μg/mL能显著抑制ICAM-1的表达。结论:三七总皂苷显著抑制炎症因子的表达。

Abstract:Objective:PNS is the main component of Xuesaitong.We select the soluble urate crystals(MSU)to induce vascular endothelial cells(HUVEC),and establish the acute gout model in vitro,and choose the vitality of HUVEC,HUVEC intercellular adhesion molecule-1(ICAM-1)expression,cell morphology as the indexes.The anti-acute gout effects of PNS were evaluated.Methods:Stimulation group was treated with final concentration 100μg/mL MSU and 0,2.5,10μg/mL PNS to deal with HUVEC for 24 hours,the viability of HUVEC was assessed by MTT test and the expression of ICAM-1 of HUVEC was detected by flow cytometry and Real-time PCR.Results:As the concentration of PNS increased,the viability of HUVEC decreased and the expression of ICAM-1 of HUVEC increased.The concentration of 2.5μg/mL PNS can significantly inhibit the expression of ICAM-1.
