中药抗复灵干预单侧输尿管梗阻大鼠肾脏病变的实验研究

目的:研究抗复灵干预单侧输尿管结扎(UUO)大鼠肾纤维化模型病变的作用。方法:健康SPF级雄性SD大鼠96只,随机分成假手术组,单侧输尿管结扎(UUO)组,抗复灵组(UUO+抗复灵),科素雅组(UUO+科素雅)分别于第7、14、21、28天各组处死6只大鼠;病理检查肾小管间质纤维化指数和免疫组化检查α-肌动蛋白(α-SMA)和COL1α2。结果:肾小管间质纤维化损害指数比较,抗复灵和氯沙坦组和模型组比较有显著性差异(P0.01)。I型胶原UUO组和假手术组比较有非常显著统计学差异(P0.01),抗复灵以及科素亚治疗组和UUO组比较有显著性差异(P0.01),并且抗复灵组在14、21、28天和氯沙坦组相比较有非常显著统计学差异(P0.01)。α-SMA有相同的变化趋势。结论:抗复灵是有效抑制肾纤维化的中药制剂。

The Role of Chinese Herbal Kangfuling on Unilateral Ureteral Obstruction Renal Lesions in Rats

Objective:To study the role of Kangfuling(KFL) on unilateral ureteral obstruction(UUO) SD rat renal fibrosis model.Methods:Healthy male SD rats SPF-class 96,were randomly divided into sham operation group,unilateral ureteral obstruction(UUO) group,Kangfuling group(UUO + KFL),Losartan group(UUO + Losartan),respectively in section 7,14,21,28 days,6 rats in each group were sacrificed.After the success of modeling test pathological examination tubulointerstitial fibrosis index and the immunohistochemical examination of α-muscle actin(α-SMA) and type 1 collagen α2(COL1α2).Results:Damage index of tubulointerstitial fibrosis in UUO group at all time points had significant differences(P<0.01).Compared KFL and Llosartan group with model group were significantly different(P<0.01).Compared I collagen of UUO group with the sham-operated group,there was a significant statistical difference(P<0.01),compared KFL and losartan treatment group with UUO group there was significant difference(P<0.01),compared the UUO KFL group 14,21,28 days with UUO Losartan group,there was a significant statistical difference(P<0.01).α-SMA has the same trends.Conclusion:KFL is effective inhibition of renal fibrosis in traditional Chinese medicine preparations.
