环维黄杨星D对高血压大鼠脑缺血再灌注突触生长素表达的影响

摘    要:
目的观察单体环维黄杨星D(CVBD)对易脑卒中型肾血管性高血压大鼠(RHRSP)脑缺血再灌注突触生长素(SYN)表达的影响。方法选择SD大鼠100只,用环形银夹使大鼠双侧肾动脉狭窄,制成RHRSP,随机分为空白组(10只)、假手术组(10只)、治疗组(40只)、对照组(40只)。后2组再用线栓法制成一侧大脑中动脉闭塞模型。免疫组织化学法、透射电镜等观察脑缺血2 h后再灌注第1、7、14、30天脑组织SYN表达。结果与空白组比较,假手术组大鼠SYN阳性表达升高,但差异无统计学意义(P>0.05)。与假手术组比较,对照组第1天SYN阳性表达明显升高,第7天达高峰,第14天开始下降(P<0.05,P<0.01),第30天则明显下降(P>0.05)。与对照组相同时间点比较,治疗组SYN阳性表达均升高(P<0.05,P<0.01)。结论CVBD促进RHRSP脑缺血损伤区神经功能的修复作用,可能与其上调脑组织SYN的表达,减轻突触超微结构损伤等机制密切相关。

Effects of CVBD on the expression of SYN in hypetensive rats with cerebral ischemia/reperfusion injury
Abstract:
Objective To observe the effects of the CVBD on the expression of synaptophysin (SYN) in stroke-prone renovascular hypertensive rats(RHRSP) after cerebral ischemia and reperfusion.Methods 100 RHRSP models were established by bilaterally narrowing the renal arteries with silver loop clips and the models of rat MCAO were created by stringing MCAO in RHRSP.They were randomly divided into blank group,sham-operation group,control group and CVBD group.The expression of SYN was observed using immunohistochemistry and transmission electron microscope at different time points at 2-h ischemia and 1,7,14 and 30-day reperfusion. Results Compared with sham operation group,the expression of SYN was increased significantly and reached the maximum after 7-day reperfusion,decreased after 14-day,and reduced markedly after 30-day in control group.The levels of the positive SYN cells in CVBD group were obviously higher than those of the control group(P<0.05,P<0.01) on d 1,d 7,d 14 and d 30 of cerebral ischemia and reperfusion(P<0.05,P<0.01).Conclusion The mechanism of CVBD to promote recovery of neural function in ischemic injury area may be related to up-regulating the expression of SYN and decreasing the ultrastructure damage of synapses.
