聚丙烯无纺布亲水性与血液相容性改性初步研究

摘    要:
目的 改善聚丙烯无纺布(PPNWF)的亲水性与血液相容性.方法 通过低温等离子体预处理的方式在PPNWF上引发接枝丙烯酸(AA)单体,制备不同AA接枝面密度的PPNWF(PP-PAA,接枝面密度分别为0.12、0.19和0.26 mg/cm2),上述PP-PAA再经EDC[1-(3-二甲氨基丙基)-3-乙基碳二亚胺]活化后,与肝素(Hep)反应,制备不同肝素结合面密度的PPNWF(PP-Hep,肝素结合面密度分别为1.78、2.34和3.21 μg/cm2);通过红外光谱、水接触角对改性材料的表面进行表征,通过溶血率、血浆蛋白吸附以及APTT试验评价其血液相容性.结果 PPNWF表面水接触角从(143±1).降到(36±5).,显示其亲水性得到明显改善;体外血液相容性评价:上述PP-PAA的溶血率分别为(4.13±0.01)%、(8.59±0.07)%和(12.78 +0.11)%,血浆蛋白吸附率分别为(7.0±1.0)%、(9.3±2.3)%和(12.0±3.1)%,APIT分别为(28.8±0.2)、(29.1±0.1)和(30.8±0.8)s;上述PP-Hep的溶血率分别为(1.08±0.01)%、(0.84±0.03)%和(1.22±0.08)%(均<5%),血浆蛋白吸附率分别为(4.8±1.1)%、(6.7±1.8)%和(4.2±1.5)%APTT分别为(28.2±1.1)、(28.9±0.2)和(28.4±0.5)s.结论 改性后的PPNWF的亲水性与血液相容性均得到明显改善.

Abstract:
Objective To improve hydrophilicity and blood compatibility of polypropylene non-woven fabrics.Methods Acrylic acid(AA) was induced grafted onto polypropylene non-woven fabric(PPNWF) via low temperature plasma pretreatment and three grafting surface density of PP-PAA were acquired,which were 0.12 mg/cm2 、0.19 mg/cm2 and 0.26 mg/cm2,respectively.The above mentioned PP-PAAs were then activated by EDC(N-ethyl-N′_[3-(dimethylamino)propyl] carbodiimide),followed by reaction with heparin,and three heparinized surface density of PP-Heps were finished,which were 1.78μg/cm2、2.34μg/cm2 and 3.21μg/cm2,respectively.The modified PPNWFs were characterized by attenuated total reflection Fourier transform infrared(ATR-FTIR) spectroscopy and a contact angle goniometer,and in vitro blood compatibility including hemolysis rate,plasma protein adsorption and APTT was investigated.Results It was found that the hydrophilicity was greatly improved according to the decrease of water contact angle from(143±1)° to(36±5)°.Hemolysis rates of the three PP-PAAs were(4.13±0.01)%、(8.59±0.07)% and(12.78±0.11)%,the adsorbent percentage of plasma protein were(7.0±1.0)%、(9.3±2.3)% and(12.0±3.1)%,and the APTT were(28.8±0.2)s、(29.1±0.1)s and(30.8±0.8)s.Hemolysis rates(all of them were less than 5%) of the three PP-Heps were(1.08±0.01)%、(0.84±0.03)% and(1.22±0.08)%,the adsorbent percentage of plasma protein were(4.8±1.1)%、(6.7±1.8)% and(4.2±1.5)%,and the APTT were(28.2±1.1)s、(28.9±0.2)s and(28.4±0.5)s.Conclusion The heparinized PPNWF had good hydrophilicity and blood compatibility.
